Insulin Production

Beginner🧪 Biotechnology & GenomicsTopic 8 of 56

Summary

Human insulin is made by bacteria carrying the human gene, replacing animal sources.

In depth

In 1978 Genentech made human insulin in E. coli; the A and B chains were produced separately and joined; now proinsulin is made in yeast or E. coli. Earlier insulin came from pigs and cattle, which caused allergic reactions.

The gene is inserted into a plasmid with a strong promoter, expressed in bacteria or yeast, purified and processed. Analogues (lispro, glargine) are engineered for faster or longer action.

Remember: the human insulin gene works in bacteria because the code is universal, but bacteria cannot splice introns so cDNA is used.
